The latter is a spice mix whereas hot Chili powder or Chili flakes is a great substitute for cayenne pepper.

Stir the seasoning for at least a minute so all the ingredients are thoroughly combined.
How to store your homemade seasoning
Put the homemade quesadilla seasoning mix in a glass jar. Store it in a dry dark place like a kitchen cabinet.
Do not put in the fridge because of condensation in the jar and the seasoning will get wet.
